ABSTRACT

Liver cancer is an important public health issue worldwide. With the improvements in high-throughput sequencing and gene editing techniques in recent years, studies have further revealed the biological mechanism of intestinal microflora in the development, progression, and metastasis of liver cancer via the gut-liver axis, and in particular, it has been found that lipopolysaccharide, a component of the outer membrane of gram-negative bacteria, can cause downstream immune cascade reactions. This article reviews the possible mechanism of action of intestinal microflora lipopolysaccharide in the development and progression of liver cancer from the aspects of the association between intestinal environmental changes and liver cancer, immunoregulation by lipopolysaccharide, and preclinical treatment.

ABSTRACT

OBJECTIVE:To in vestigate the effects of dexmedetomidine (Dex)on SIRT 1/Akt/GSK3β/β-catenin signaling pathway in cerebral injury of sepsis model rats ,and explore the mechanism of its protecitve effect on cerebral injury. METHODS : A total of 80 male SD rats were randomly divided into sham operation group (Sham group ),sepsis group (CLP group ),CLP+Dex group(10 μg/kg Dex),CLP+Dex+Sirtinol group (10 μg/kg Dex+2 μL/100 g SIRT 1 inhibitor sirtinol ),with 20 mice in each group. Two hours before modeling ,CLP+Dex+Sirtinol group was injected with sirtinol via lateral ventricle. Sepsis model was induced by cecal ligation and perforation in each group (in sham group ,only operation was performed but no ligation was performed). At 0,3,6 h after modeling ,CLP+Dex group and CLP+Dex+Sirtinol group were given Dex (10 μg/kg) intraperitoneally,Sham group and CLP group were given constant volume of normal saline intraperitoneally. Cerebral tissue water content,Evans blue (EB)content, apoptosis in cerebral cortex ,the levels of IL- 1β and TNF-α in cerebral tissue as well as the protein expression of SIRT 1,p-Akt,p-GSK3β and β-catenin in hippocampus were detected 24 h after last medication. RESULTS : Compared with Sham group ,cerebral tissue water content ,EB content ,the number of apoptotic cells in cerebral cortex as well as the levels of IL- 1β and TNF-α in cerebral tissue were increased significantly(P<0.05),while the protein expression of SIRT 1, p-Akt,p-GSK3β and β-catenin in hippocampus were decreased significantly (P<0.05). Compared with CLP group ,cerebral tissue water content ,EB content ,the number of apoptotic cells in cerebral cortex as well as the levels of IL- 1β and TNF-α in cerebral tissue were decreased significantly in CLP+Dex group (P<0.05),while the protein expression of SIRT 1,p-Akt,p-GSK3β and β-catenin in hippocampus were increased significantly (P<0.05). Sirtinol could significantly reverse the above-mentioned cerebral protection and factor regulation effects of Dex (P<0.05). CONCLUSIONS :Dex can protect the cerebral tissue of sepsis model rats, which may play an anti-inflammatory and anti-apoptotic role by activating SIRT 1/Akt/GSK3β/β-catenin signaling pathway ,so as to reduce cerebral edema ,protect blood-brain barrier and reduce cerebral injury.

ABSTRACT

Purpose To evaluate the value ofintravoxel incoherent motion (IVIM) imaging in diagnosis of liver fibrosis staging in rats.Materials and Methods Rabbit models of liver fibrosis at different stages were established. All rabbits were divided into four groups based on the pathological results of fibrosis grading as S1-S4.The 1VIM imagings with 8 b-values (0,50,100,200,300,800,1000,1200 s/mm2) were performed. The diffusion coefficient (D),perfusion-related coefficient (D*),and perfusion fraction (f) were calculated and compared between control (only injection of saline) and S 1 group,S2 and S3 group. Results The D value was significantly lower in S1 group compared with control group (P<0.05),but the D* and f values showed no significant difference between the two groups (both P>0.05).With the progression of liver fibrosis, the D,D* and f value decreased gradually;the D* value showed significant difference between S2 and S3 group (P<0.05),but the D and f values showed no significant differences between the two groups (both P>0.05).Conclusion The D value is useful for differentiation of normal liver and hepatic fibrosis of S1 stage, while the D* is valuable for differentiation of hepatic fibrosis of S2 and S3 stage. However, the f value neither could detect early fibrosis, nor could differentiate hepatic fibrosis staging.IVIM imaging provides a noninvasive method for early and accurate staging of liver fibrosis, which may be of great help in clinical diagnosis and treatment.

ABSTRACT

[ ABSTRACT] AIM:To investigate the effect of polysaccharide from Fructus corni ( PFC) on cardiomyocytes against hypoxia/reoxygenation ( H/R) injury and its possible relationship with ROS/PKC/p38 MAPK pathway.METHODS:Prima-ry cardiomyocytes were isolated from neonatal SD rats and randomly divided into normal group, H/R group, PFC (20 mg/L, 100 mg/L and 200 mg/L) preconditioning+H/R groups, chelerythrine+PFC (100 mg/L)+H/R group and SB203580+PFC (100 mg/L)+H/R group. The cell viability was measured by inverted microscopic observation.Apoptosis in the car-diomyocytes was detected by Hoechst 33258 staining and fluorescence microscopy. The levels of lactate dehydrogenase ( LDH) and superoxide dismutase ( SOD) in the cell culture supernatants, and the reactive oxygen species ( ROS) in the cells were also measured by microplate reader. The protein levels of PKC, p-p38 MAPK and HSP70 in the cells were detec-ted by Western blotting.RESULTS:Compared with normal group, the cell viability and beating frequency were decreased in H/R group.LDH and ROS contents, apoptotic rate and p-p38 MAPK level increased significantly (P<0.01).Compared with H/R group, PFC preconditioning increased beating frequency, SOD activity and the protein level of PKC and HSP70, and decreased ROS production, the protein level of p-p38 MAPK and cell apoptotic rate. However, the effect of PFC was in-hibited by chelerythrine or SB203580.CONCLUSION:PFC may protect cardiomyocytes from hypoxia/reoxygenation injury. Its mechanism is possibly involved in the inhibition of ROS via increasing the activity of SOD and the activation of PKC, and suppression of excessive activation of p38 MAPK.

ABSTRACT

BACKGROUND: Long term taking cyclosporin A(CsA)can inhibit osteoblastic differentiation and induce osteoporosis.Equol, which has greater binding affinity to estrogen receptors, can stimulate the proliferation and osteoblastic differentiation.OBJECTIVE: To investigate whether equol may protect against the proliferation and osteoblastic differentiation inhibited by CsA in mouse bone marrow mesenchymal stem cells(BMSCs)cultures and analyze signal pathway of protection.METHODS: Primary mouse BMSCs were cultured by using attachment method and assigned to five groups, which respectively treated with equol or/and CsA in the presence or absence of ICI182780,an estrogen receptor antagonist, and Nω-nitro-L-arginine methyl ester.Under an inverted microscope, morphological changes and mineralization ability of BMSCs were observed. The cell proliferation was measured by[3H]-thymidine incorporation. The osteoblastic differentiation and mineralization of extracellular matrix in BMSCs was assessed by measuring alkaline phosphatase activity and calcium deposition, respectively.Nitric oxide production in the conditioned media and cyclic guanosine monophosphata(cGMP)content in BMSCs were determined by using commercial nitric oxide and cGMP kit, respectively.RESULTS AND CONCLUSION: Equol reversed the decreased[3H]thymidine incorporation(P < 0.05),alkaline phosphatase activity(P < 0.05)and calcium deposition(P < 0.01)of CsA,which was accompanied with the changes of nitric oxide production(P < 0.01)and cGMP content(P < 0.01).The group by co-treatment with equol and CsA possessed higher cells growth density and small mineralized nodes than CsA group on day 12 under an inverted microscope. Moreover, the equol-reversed effect was abolished by ICI182780 and Nω-nitro-L-arginine methyl ester. These indicated that equol can reverse the inhibition of CsA on the proliferation and osteoblastic differentiation of mouse BMSCs through estrogen receptor/nitric oxide/cGMP signal pathway.

ABSTRACT

MicroRNAs (miRNAs or miRs) are a class of small non-protein-coding RNAs, approximately 18 to 25 nt long. MicroRNAs can act as endogenous RNA interference. MicroRNAs can posttranscriptionally regulate the expression of hundreds of their target genes, controlling a wide range of biological functions such as cellular proliferation, differentiation, and apoptosis. MicroRNAs can posttranscdptionally regulate the expression of genes by hybridizing to complementary sequences in 3' UTR (3' untranslated region) of target messenger RNA (mRNA), repressing the translation of mRNA or increasing the instability of mRNA. A number of miRNAs are mapped to cancer-associated fragile regions (FRAs) as well as in minimal regions of loss of heterozygosity, minimal regions of amplification, or common breakpoint regions in the genome, suggesting that miRNAs might be involved in tumorigenesis. Many miRNAs are up or down-regulated in cancers as potential oncogene or tumor suppressors. It has been considered that the mutation of a series of oncogene or anti-oncogene gradually causes tumorigenesis. The conventional points of view were changed with the finding of non-protein-coding RNAs. MiRNA as members of non-protein-coding RNAs may play an important role in regulating tumor formation. Recent studies have in-vestigated the relationship of miRNAs with neoplasia, development, treatment and prognosis. Lung cancer is the leading cause of cancer-related deaths all over the world. Its etiology is primarily genetic and epigenetic damage caused by tobacco smoke. Systematic analysis of RNA and protein expression levels of thousands of genes has also contributed to defining the molecular net work of lung carcinogenesis. MiRNAs are closely related to lung cancer and play an important part in the diagnosis, therapy, surveillance and prognosis of lung cancer. We reviewed some miRNAs closely associated with lung cancer such as miRNA-126, miRNA-221, miRNA-222, has-mir-221, a polycistronic microRNA cluster miR-17-92, miRNA-128b, has-mir-137, has-mir-182, has-mir-372 and miRNA let-7. We summarized the roles of miRNAs in the genetic susceptibility, invasion or metastasis, diagnosis, therapy and prognosis of lung cancer.

ABSTRACT

Two pairs of primer targeting to the genes encoding the major outer membrane protein (MOMP) and the polymorphic membrane protein (PMP) of Chlamydophila psittaci (Cps) designed and used in the fluorescent quantitative PCR assay in the detection of this microorganism were compared in the their sensitivity and specificity. It was demonstrated that both pairs of primer could be used successfully to detect the presence of Cps of the epidemic strains isolated in China. The specificity of PMP primer used was higher than that of MOMP primer. In case of high target concentration in samples, the sensitivity of the former was also superior to that of the latter, but the amplification efficiency of the former was lower than that of the latter. The MOMP primer seemed to be more suitable to detect Cps by using the real-time quantitative assay. As demonstrated by the real-time quantitative PCR assay, great difference in the genomic copy numbers of the PMP gene existed in the epidemic strains of Cps isolated in China.

ABSTRACT

The aim of the present studies was to investigate the cardioprotection of late IP at 72 h and determine the involvement of iNOS, nNOS and COX-2 in this protection. Conscious rabbits were preconditioned with three cycles of 5-minute coronary occlusion/5-minute reperfusion. The myocardial infarct area in the rabbits preconditioned 72 h earlier was significantly smaller than that in control rabbits. The activity of lactate dehydrogenase (LDH) and the level of 6-Keto-PGF1alpha in the rabbits preconditioned 72 h earlier were lower than those in control rabbits. The left ventricular systolic pressure (LVSP) and maximal velocity of contraction and relaxation (+/- dP/dtmax) were improved in rabbits preconditioned 72 h earlier. The nNOS-selective inhibitors N-propyl-L-arginine and selective cyclooxygenase-2 (COX-2) inhibitor celecoxib completely blocked the protection of late IP at 72 h, whereas the iNOS selective inhibitor S-methybisothiourea had no effect. In conclusion, the cardioprotection observed in the final stage of late IP (72 hours) is mediated by nNOS or COX-2, but not by iNOS.

ABSTRACT

To study the effect of soybean isoflavone on liver oxidative stress resulting from 60Co-gamma rays. Methods: Totally 80 normal female Kunming mice were evenly randomized into 5 groups according to body weight: 3 intervention groups, single irradiation group and normal control group. The normal group and single irradiation groups were given 0.5% CMC-Na, and the 3 intervention groups were given different doses of soybean isoflavone (50 mg/kg, 100 mg/kg, 400 mg/kg) respectively for 14 d. The whole body of single irradiation group and intervention groups were subjected to 4.56 Gy 60Co-γ radiation once on the 7th day, and then the mice were killed on the 2nd day and the 7th day after radiation. Results: The CAT activity of liver tissue of 100,400 mg/kg intervention groups and 3 SI groups were significantly increased on the 2nd day and 7th day after irradiation(P<0.05), respectively; the GSH-Px activity of 100 mg/kg SI group was significantly increased(P<0.05) on the 7th day after irradiation; the T-SOD activity of 50 mg/kg SI group was significantly decreased (P<0.05) on the 2nd day after irradiation, while no difference was observed among remaining groups. The MDA content of 100 mg/kg group was significantly decreased on the 7th day after radiation compared with control group, and MDA content of each group subjected to irradiation were increased on the 2nd day after irradiation, but 3 SI groups nearly decreased to normal level on the 7th day after irradiation. Conclusion: The soybean isoflavone can enhance the antioxidant capability of mice, but it does not show a dose-effect relationship.
